Abstract Objective To investigate the effects of Chinese medicine Danlou tablets and Xuefu Zhuyu granules on blood lipid metabolism and inflammatory reaction in rat model of atherosclerosis. Methods Forty male Wistar rats were randomly divided into five groups, namely normal control group, model control group, atorvastatin group, Danlou group and Xuefu Zhuyu group. Normal control group was given basic forage, while other four groups were given high fat forage plus intraperitoneal injection of vitamin D3 to establish atherosclerosis model, and then received intragastric administration of saline, atorvastatin suspension, Danlou tablet suspension and Xuefu Zhuyu granule suspension for 8 wk, respectively. After intervention, serum lipid and IL- 6, TNF- α, MCP- 1, ox- LDL, LP- PLA2 levels were measured by ELISA method, the expression of IL- 6, TNF- α, MCP- 1, LP- PLA2 mRNAs in thoracic aorta were detected by real- time fluorescent quantitative PCR (RT- fqPCR). Results Compared with normal control group, serum TC, TG, LDL- C, IL- 6, TNF- α, MCP- 1, ox- LDL and LP- PLA2 levels and the expressions of IL- 6, TNF- α, MCP- 1 and LP- PLA2 mRNAs in thoracic aorta of model group were significantly increased (al P<0.05). Compared with model control group, serum TC, TG, LDL- C, IL- 6, TNF- α, MCP- 1, ox- LDL and LP- PLA2 levels and the expressions of IL- 6, TNF- α, MCP- 1 and LP- PLA2 mRNAs in thoracic aorta of atorvastatin group and Danlou group were significantly decreased (al P<0.05);and there were no significant differences between atorvastatin group and Danlou group (P >0.05). Serum TC, IL- 6, TNF- α, MCP- 1 levels and expressions of IL- 6, TNF- α, MCP- 1 mRNA in thoracic aorta of Xuefu Zhuyu group were significant decreased (al P<0.05) compared with model control group, while there were no significant differences in serum TG, LDL- C, ox- LDL, LP- PLA2 levels and expression of LP- PLA2 mRNA in thoracic aorta between two groups (P>0.05). Conclusion Both Danlou tablets and Xuefu Zhuyu granules can inhibit expression of inflammatory factors and relieve inflammatory reaction. Danlou tablets showed a stronger effect in regulating lipid metabolism than Xuefu Zhuyu granules.
