Abstract Objective To investigate the effects of Danlou tablets on the vascular endothelial function and the expression of LOX- 1 in atherosclerotic rats.Methods Forty male Wistar rats were randomly and equally divided into five groups,namely control group,model group,atorvastatin group(ATV group),low- dose of Danlou group(DLD group),and high- dose of Danlou group (DLG group).Control group fed basic diet.Other four groups fed high fat diet plus intraperitoneal injection of vitamin D3 to create atherosclerosis model,and then received intragastric administration of saline,atorvastatin,low dose of Danlou tablets and high dose of Danlou tablets for 8 weeks,respectively.Serum lipid,ET,NO and ox- LDL levels were detected by ELISA method,the protein and gene expression of1ectiu- like oxidized low- density lipoprotein receptor- 1 (LOX- 1) in thoracic aorta were measured by Western- blot and real- time fluorescent quantitative PCR.Results Serum TC、TG、LDL- C、ET and ox- LDL levels,and LOX- 1 in thoracic aorta were significantly higher (P<0.05)and serum NO level was significantly lower (P<0.05)in model group than in control group (al P<0.05).Serum TC、TG、LDL- C、ET and ox- LDL levels (P<0.05)as well as LOX- 1 in thoracic aorta were significantly lower,and serum NO level was significantly higher in ATV group、DLD group and DLG group than in model group (all P<0.05).There was no significant difference of these parameters between ATV group and DLD group or DLG group.Conclusion Danlou tablets is effective in improving vascular endothelial function of athrosclerotic rats,which may be due to the decrease of serum ox- LDL level and LOX- 1 expression.
