Abstract Objective To observe therapy effect of Salvianolic acid (SA) combined with amlodipine on rats with hypertension and hyperlipidemia. Methods 24 spontaneously hypertensive rats (SHR) fed high- fat diet for 2 weeks and then randomly fed amlodipine (AMl group, n=6), amlodipine plus atorvastatin (AMLAT group, n=6), SA plus amlodipine (SAAML group, n=6) or water (control group, n=6), respectively, as well as high- fat diet for 4 weeks. Blood pressure, serum lipids, superoxide dismutase (SOD) and malondialdehyde (MDA) of plasma, liver and myocardium were measured before and after treatment. Results After 4 weeks treatment, blood pressure increased and HDL- C decreased significantly in control group(P<0.05or0.01). Serum TG, LDL- C, LDL- C/HDL- C, hepatic TC and TG decreased, plasma and myocardial MDA decreased, SOD and ratio of SOD/MDA increased significantly in SAAML group compared to control group ( al P<0.05 or 0.01). Blood pressure, LDL- C, LDL- C /HDL- C, hepatic TC, myocardial MDA decreased, myocardial SOD/MDA increased significantly in AML and AMLAT groups compared to control group(P<0.05 or 0.01). Hepatic TG was significantly lower in AMLAT group than control group (P<0.05). Conclusion Salvianolic acid combined with amlodipine may have synergetic effects on blood pressure and lipid lowering in SHR with hyperlipidemia. The possible mechanisms are related to inhibition of lipid peroxidation and protection of endothelial function.
