Abstract Objective To explore the changes of matrix metalloproteinase(MMP)- 2, 9 in left ventricle (LV)of rats with diastolic heart failure. Methods Thirty- four male Sprague- Dawley rats were randomly divided into normal control group (n=17)and model group (n=17). One renal artery was ligated in model group and sham operation was done in control group. 12 weeks later,LV hemodynamics was determined by a cardiac catheter and the ratio of LV mass (LVM) to body weight(BW) was measured. MMP- 2, 9 of LV myocardium were measured by enzyme linked immunosorbent assay(ELISA) and immunohistochemistry(IHC). Results The systolic blood pressure and carotid artery pressure increased by 24.69%and 32.68%(al P<0.01), LVM/BW and LV end diastolic pressure increased by 38.38% and 418.56%(P<0.05 or 0.01),+dp/dt max increased by 8.56%(P<0.05), and- dp/dt max decreased by 11.34%(P<0.01) in model group than in control group. The expression of MMP- 2、9 of LV myocardium increased by 29.8% and 7.95% on ELISA or by 132.61%and 29.71%on IHC, respectively, in model group than in control group (al P<0.01). Conclusion MMP- 2、9 synthesis increases in LV myocardium of rats with diastolic heart failure.
